Vildagliptin is a DPP-4 inhibitor administered twice daily. Weight neutral with low hypoglycaemia risk.
Also known as: Galvus
Inhibits DPP-4, increasing incretins. Reversible activity (unlike sitagliptin).
Reversibly blocks DPP-4, increasing GLP-1 and GIP. Reversible binding allows residual activity even at low drug levels.
Oral bioavailability: 85%. Food does not significantly affect absorption. Tmax: 1-2 hours.
Hydrolytically metabolised by peptidase in erythrocytes (inactive Ba41-795L metabolite). Does not interfere with CYP.
~2 hours (vildagliptin); ~3 hours (Ba41-795L). Elimination: ~70% urine (metabolites).
