Competitive and reversible inhibitor of HMG-CoA reductase, the rate-limiting enzyme in hepatic cholesterol synthesis. High-intensity statin.
Selectively inhibits HMG-CoA reductase, blocking conversion of HMG-CoA to mevalonate, a cholesterol precursor. Upregulates hepatic LDL receptors.
Oral bioavailability: ~20%. Food does not significantly affect absorption. Tmax: 3-5 hours.
Minimally metabolised by CYP2C9 (10%). Not a prodrug. Slightly inhibited by cimetidine. Low CYP-mediated interaction potential.
~19 hours. Elimination: ~90% faeces, ~10% urine. Long half-life allows once-daily dosing.
